Students need to be well aware of the syllabus in order to score well in the ISC Class 12 Biotechnology examination. Biotechnology aims to enable candidates to develop an understanding of how materials are provided by biological agents to provide goods and services, to appreciate the role of Biotechnology in improving health care, the contribution of Biotechnology to human welfare, and to develop scientific attitude towards biological phenomena.

 

Paper 1. Theory - 70 Marks

1. Molecular Biology 

This part requires an understanding of nucleic acids, their estimation and biochemical structure. It elaborates on techniques of nucleic acid estimation, protein synthesis,  concept of reverse transcription, properties of genetic code, translation of RNA to protein and the Operon concept.

2. Genetic Engineering

This part concerns Gene cloning and genetic engineering, the concept of cloning and vectors, innovations in Biotechnology and Gene analysis techniques.

3. Cell Culture Technology

General tools and techniques involved in cell culture technology and their applications are discussed here. Further discourse is related to Microbial Culture and its application, Plant Tissue Culture and its application, Animal Cell Culture and its application.

4. Bioinformatics

Definition and need of bioinformatics are talked about here, besides an introduction to global bioinformatics databases and data retrieval tools (ENTREZ, Taxonomy Browser),  definition of genomics, tools and applications and types of Proteomics. 

 

Paper 2. 

Practical Work - 15 Marks

A list of experiments to be completed by the candidates:

  1. Paper Chromatography

  2. Preparation of Buffers

  3. Preparation of Culture Media

  4. Sterilization of culture medium and other materials

  5. Preparation of various forms of culture media

  6. Inoculation and incubation of Lactobacillus on the culture medium in Petri plate

  7. Identification of bacteria by Gram +ve and Gram -ve

  8. Action of enzymes on starch

  9. Isolation of DNA from plants

  10. DNA estimation by colorimeter (DPA method)

  11. Protein estimation by colour reaction (Bradford Test)

  12. Cell viability test (Evan’s blue dye)

  13. Isolation of milk protein

  14. Chromatography to find adulteration in species

  15. Demonstration of cell counting by haemocytometer

  16. Experiment to show Saponification

 

Project Work - 10 Marks

Candidates are assessed by a visiting examiner on creatively executing one project/assignment on an aspect of Biotechnology. Reports should be kept neat and elegant and no extra credits are given for type-written material/decorative cover, etc.


Practical File - 5 Marks

The Visiting Examiner assesses students on the basis of the practical file maintained by them during the academic year.
