A prokaryotic cell consists of a chromosome that is circular and is very large in length as compared to the other cell dimensions which are compressed to get fit in the cell. The nucleoid is made up of the formation of chromosomal architectural proteins and RNA molecules which are the content creation and functional arrangement and DNA supercoiling. A single cell constitutes multiple copies and consists of a widely varied length of a genome. In Escherichia coli the negatively supercoiled chromosomal DNA gets folded into plectonemic loops and gets diffused into each other in different physical regions. It does not contain any structure of a bacterial nucleoid but consists of features in it. The loops are organized spatially which are known as macrodomains and are found in the DNA sites which are interacted frequently and the interactions are there between them.
