Hint: We need to know that the stain can be easily distinguished from the material, medium or the surface and it is a discoloration found on the surface. The stain is generated by the physical interaction or a chemical interaction between two dissimilar materials. And there are different techniques used for the removal of stains. This stain needs both physical treatment and chemical treatment.

We have to know that the staining is a method which is used to strengthen contrast in the samples normally at the microscopic sizes. Generally the stains will enhance the visualization of the cellular components by using a microscope. The staining is mainly divided into four types and that is, simple staining, gram staining, acid fast staining, endospore staining.
The simple stain is mainly used to find out the cell size, arrangement and cell shape and it contains only one stain which readily accepts a hydrogen ion or gives a hydroxide ion. And the stain becomes positively charged.
Gram staining is a technique which is used for differentiation of physical and chemical properties of cell walls of bacteria.
The acid fast stain is used to determine the infected body substance, blood or tissues by bacteria.
The endospore staining is used to identify the bacterial endospores.

The stainings are mainly used to differentiate between dead cells and live cells which are present in the sample. And some procedure is followed for the staining process. In the first step, the cells are stained by using the crystal violet and adding some setting agent. Next the alcohol is applied. And this will selectively separate the stain. At last, a secondary stain is added which decolorizes the cell pink.
