EcoRI is a restriction enzyme isolated from the E.coli bacteria in which the R represents the particular strain named RY13 and I represent that it was the first enzyme isolated from this strain. This enzyme cleaves DNA double helix into fragments at some specific sites.

Additional Information: The EcoRI is a restriction enzyme that creates four nucleotide sticky ends with the end of 5'. The enzyme cuts at the recognition site of G/AATTC which has a complementary sequence of CTTAA/G. Here "/" represents the phosphodiester bond that the enzyme breaks in the DNA molecule.
This enzyme is useful for many molecular genetic techniques such as cloning, DNA screening, and deleting sections of DNA in vitro. It is used in the cutting DNA prior to ligation because of its property of generating sticky ends of DNA which makes the ligation reaction more efficient. It can also carry out star activity i.e. the non-site-specific cutting. But this process depends on several factors such as low salt concentration, high glycerol concentration, an excessive amount of enzyme, high pH, etc. The cut produces sticky ends on the vector mainly viral DNA or plasmids.

note: Like many restriction endonucleases, in the primary structure of EcoRI, it contains the PD..D/EXK motif. It is a humid er of a 31 kilodalton subunit which consists of a globular domain of the α/β architecture. It has been co-crystallized with the sequence it normally cuts and this structure shows that the subunits of the enzyme homodimer interact with DNA symmetrically. The two α-helices from the subunits together form the four-helix bundle.
